Virtsan bakteeriviljely tutuksi : oppimateriaali virtsan bakteeriviljelystä Seinäjoen keskussairaalan mikrobiologian laboratorioon

Abstract

Virtsatieinfektiot ovat hengitystieinfektioiden jälkeen toiseksi yleisimpiä infektioita Suomessa. Vuosittain hoidetaan satojatuhansia virtsatieinfektioita. Tauti todetaan oireiden, liuskapikatestien ja bakteeriviljelyn perusteella. Bakteeriviljelyssä bakteeri viljellään maljan kiinteälle agarpohjaiselle elatusaineelle. Yksi lisääntymiskykyinen bakteeri jakaantuu yön yli tapahtuvassa kasvatuksessa miljooniksi jälkeläisiksi ja muodostaa maljalle silmillä nähtäviä pesäkkeitä. Eri bakteerilajit muodostavat erilaisia pesäkkeitä. Näistä pesäkkeistä voidaan tehdä erilaisia tunnistustestejä, jotka perustuvat bakteerin biokemiallisiin ominaisuuksiin.

Tämän opinnäytetyön tarkoituksena oli valmistaa selkeä ja kattava oppimateriaali virtsan bakteeriviljelystä Seinäjoen keskussairaalan kliinisen mikrobiologian osastolle. Oppimateriaali on suunnattu pääasiassa harjoitteluun tuleville opiskelijoille ja uusille työntekijöille, mutta on myös hyvä kertausmateriaali pitkään poissa olleille työntekijöille. Oppimateriaalissa käytettiin runsaasti havainnollistavia kuvia virtsan bakteeriviljelyn suorittamisen menetelmistä ja virtsatieinfektioita aiheuttavien bakteerien pesäkkeistä viljelymaljoilla. Tämän opinnäytetyön tavoitteena oli valmistaa yhtenäinen ja selkeä oppimateriaali, joka nopeuttaisi ja selkeyttäisi asioiden oppimista virtsan bakteeriviljelyprosessista.

Opinnäytetyö toteutettiin toimeksiantosopimuksen luvalla Seinäjoen keskussairaalan mikrobiologian osaston kanssa. Tämä opinnäytetyö on toiminallinen kehittämistyö, koska tuotoksena syntyi oppimateriaali ja prosessista tuotettiin raportti. Opinnäytetyö toteutettiin Turun ammattikorkeakoulun opinnäytetyöohjeiden mukaan ja noudattaen hyvää tieteellistä käytäntöä. Lähdekritiikkiin kiinnitettiin tarkkaa huomiota ja lähteet merkittiin tarkasti. Eettisiä ongelmia opinnäytetyössä ei ollut, koska tutkimuksen kohteena olivat ainoastaan virtsan bakteeriviljelymaljat. Maljat kuvattiin niin, että näytteenantajaa ei ole mahdollista selvittää.

Tuotoksena valmistui kirjallinen oppimateriaali Virtsan bakteeriviljely tutuksi-oppimateriaali Seinäjoen keskussairaalan mikrobiologian osastolle. Sivuja tuotoksessa on 50 ja kuvia 30 kappaletta. Tutkimustehtävä suoritettiin onnistuneesti ja tuotoksena syntyi selkeä ja laadukas oppimateriaali, jota voidaan tulevaisuudessa hyödyntää virtsan bakteeriviljelyn opetuksessa Seinäjoen keskussairaalan mikrobiologian laboratoriossa.

In Finland urinary tract infections are the second common infections, as the respiratory infections are the most common infections. Yearly hundreds of thousands urinary tract infections are being treated in Finnish hospitals and health care centers. Urinary tract infections can be identified by analysis of the symptoms, rapid tests or with the bacterial culture from patients` urine sample. In bacterial culture bacteria is set to grow plate on a solid agar-based growth medium. One living bacteria can produce millions of offspring when it’s set to grow overnight. These bacteria will form visible colonies. Every bacterium will create a different kind of colony and with these colonies it’s possible to perform a series of identification tests that are based on the biochemical characteristics of the bacteria.

The objective of this work was to make a simple and comprehensive guidebook of urine cultures culture for Clinical Microbiology Laboratory of Seinäjoki Central Hospital. The material is directed for the students and for the new employees, but it also provides a good informative material for employees that have been absent for a long time. Thesis was built together by using lots of illustrative pictures of urine cultures and colonies of urinary tract infection causing bacteria on culture plates the main objective of this thesis was to create a clear and consistent guidebook that would educate and aid the process of urine culture.

This work is done with the an agreement of the Clinical Microbiology Laboratory department of the Seinäjoki central hospital. The described work is a functional development of the urethral bacterial culture process and as the result of this assignment came the guidebook of the process and also literal report was created of carrying out the assignment. The work was made to meet the standards set by the University of Applied Sciences of Turku and using appropriate scientific methods. Much attention was used for investigating the reliability of the sources and they were listed thoroughly. During the project there were no ethical problems, as the study was based on the information that was gathered from culture plates that were named with a serial number.

As the result of this project came the literal guidebook ‘’Virtsan bakteeri-viljely tutuksi’’- The Guidebook of urine culture for the microbiological department of Seinäjoki Central Hospital. The guidebook consists of 50 pages and 30 illustrative pictures. The assignment was carried out successfully and as the outcome came a high-quality guidebook that can be used in education of urine culture at the laboratory of microbiological Clinical Microbiology Laboratory of Seinäjoki Central Hospital.
