| dc.description.abstract | Opinnäytetyön tavoitteena oli tutkia viiden erilaisen leimareagenssin (luminoivien lantanidikelaattien) toimivuutta NT-proBNP-immunomäärityksessä sekä niiden ominaisuuksien vaikutusta toimivuuteen.
Leimareagenssit konjugoitiin vasta-aineeseen ja ne testattiin NT-proBNP-immunomäärityksessä sekä käsin tehtävällä määrityksellä että kuivattuna AQT90 FLEX -immunoanalysaattorilla. Leimatuista vasta-aineista mitattiin ja laskettiin kelaattien fotofysikaalisia ominaisuuksia.
Tutkimuksen tuloksena on referenssiä neljä kertaa kirkkaampi Europium-kelaatti. Sen toimivuutta todennettiin NT-proBNP-vasta-aineen lisäksi kahdella muulla analyytillä. Moninkertainen kirkkaus tarkoittaa korkeampaa signaalitasoa, jota voidaan hyödyntää määrityksen herkkyyden nostamisessa. | - |
| dc.description.abstract | The aim of the thesis was to study the performance of five different labeling reagents (luminescent lanthanide chelates) in the NT-proBNP immunoassay and the effect of chelate properties on functionality.
The labeling reagents were conjugated to the antibody and were tested in the NT-proBNP immunoassay both manual assay and dried with AQT90 FLEX immunoanalyzator. The photo-physical properties of labeling reagents were measured and calculated from labeled antibodies.
The result of this study is a four-fold brighter Europium chelate than reference chelate. Its performance was verified with two other analytes. Higher brightness means a higher signal level that can be utilized to increase the sensitivity of the assay. | - |
