Tiivistelmä
Tabletti on tunnetuin ja eniten käytetty lääkemuoto. Tablettien suosio perustuu vaivattomaan käsittelyyn ja varastoitavuuteen, annoksen tarkkuuteen, tuotteen säilyvyyteen sekä asiakkaan näkökulmasta käytön helppouteen.
Tablettien yhtenä laatuvaatimuksena on mekaaninen kestävyys. Tämän opinnäytetyön tarkoituksena oli tutkia kolmen tablettimuotoisen lääkkeen muodon muuttamista ja muutoksen vaikutusta lääkkeiden kestävyyteen. Tavoitteena oli estää tuotteiden katkeaminen valmistusprosessin ja pakkaamisen aikana.
Sopivaa muotoa valikoitaessa tehtiin useita tutkimuksia aloittaen pienen mittakaavan esitestistä. Tutkimusten edetessä siirryttiin tuotantomittakaavan koe-erien valmistukseen ja lopuksi tehtiin kaikkien kolmen tutkittavan tuotteen prospektiivinen prosessivalidointi valikoiduilla tablettimuodoilla. Tutkimuksissa valmistettuja tabletteja testattiin laajasti. Orionin tablettivalmistusosastolla tehtiin prosessikontrolleja valmistuksen aikana ja yrityksen omissa laboratorioissa analysoitiin valmistettuja tabletteja tablettimuodon valinnan tueksi ja laadun varmistamiseksi.
Opinnäytetyön tuloksena muutettiin kahden tuotteen muotoa. Tuotteet ovat olleet muutoksen jälkeen mekaanisesti kestävämpiä eikä katkeaminen ole aiheuttanut ongelmia valmistus- ja pakkausprosessissa. Tuloksena saavutettiin sujuvampi ja tehokkaampi valmistusprosessi, joka minimoi riskejä sekä parantaa tuotteiden laatua ja kannattavuutta. The tablet is the best known and most widely used dosage form. The popularity of the tablets is based on easy handling and storability, dose accuracy, shelf life of the product and the ease of use from the customer's point of view.
One of the quality requirements for tablets is its mechanical sustainability. The purpose of the present Master’s thesis was to investigate the shape change of three tablet-shaped drugs and the effect of the change on the sustainability of drugs. The aim was to prevent the products from breaking during the manufacturing process and packaging.
When selecting the appropriate format, several studies were conducted, starting with a smallscale pre-test. As the studies progressed, production-scale test batches were developed and finally a prospective process validation of all three investigated products with the selected tablet forms was performed. The tablets prepared in the studies were extensively tested. The process controls were performed at Orion's tablet manufacturing department during manufacturing and the manufactured tablets were analyzed in the company's own laboratories to support the choice of tablet form and to ensure quality.
As a result of the thesis, the shape of the two products was changed. The products have been more mechanically sustainable since the change and the breakage has not caused any problems in the manufacturing and packaging processes. The result was a smoother and more efficient manufacturing process that minimizes the risks and improves the product quality and profitability.
