Tiivistelmä
Entsyymit katalysoivat eli nopeuttavat kemiallisia reaktioita ja ovat välttämättömiä solujen toiminnalle. Ne ovat lähes aina proteiineja ja niiden oikea kolmiulotteinen rakenne mahdollistaa niiden biologisen aktiivisuuden. Entsyymien toimintaa voidaan estää erilaisten estäjien eli inhibiittoreiden avulla, joita ovat useat lääkeaineet, ja moni entsyymi on myös aktiivisen lääkekehityksen kohteena.
Hydroksisteroidi-17-beta-dehydrogenaasi tyyppi 10 (HSD17β10) -geenin aktivoitumisen tuloksena syntyy HSD17β10-entsyymiä, joka tunnetaan myös tyypin II 3-hydroksiasyyli-CoA-dehydrogenaasina. Tämä mitokondrionaalinen entsyymi saa aikaan rasvahappojen, alkoholien ja steroidien hapettumista. Se pystyy katalysoimaan esimerkiksi biologisesti aktiivisen estrogeenin, 17β-estradiolin, hapettumista vähemmän aktiiviseksi estroniksi, 3α-androstanediolin hapettumista 5α-dihydrotestosteroniksi ja allopregnanolonin hapettumista 5α-dihydroprogesteroniksi.
HSD17β10-entsyymiaktiivisuutta on havaittu aivojen eri osa-alueilla, eniten hippokampuksessa. Alzheimerin tautia sairastavilla potilailla HSD17β10-entsyymin tasot kohoavat, jolloin hermostoa suojaavien estradiolin ja allopregnanolonin tasot vastaavasti laskevat. HSD17β10-entsyymin inhibitiolla on mahdollista pitää yllä aivojen hermosoluja suojaavia steroiditasoja, minkä vuoksi tämä entsyymi onkin mielenkiintoinen lääkekehityksen kohde.
Opinnäytetyön tavoitteena oli kehittää uusi HSD17β10-entsyymin aktiivisuustesti, jota voitaisiin myöhemmin käyttää lääkeainekandidaattien seulonnassa. Menetelmän kehityksessä hyödynnettiin tieteellisissä julkaisuissa olevaa tietoa ja yrityksessä olevaa kokemusta vastaavista entsyymiaktiivisuustestien kehittämisestä. Työhön sisältyi entsyymin toimintaan vaikuttavien tekijöiden analyysejä, jotka toteutettiin 96-kuoppalevyformaatissa fluoresenssi- ja UPLC-menetelmillä.
Opinnäytetyölle asetetut tavoitteet toteutuivat, sillä uusi HSD17β10-entsyymin aktiivisuustesti saatiin kehitettyä osaksi yrityksen lääkeaineseulontaa. Enzymes catalyze or accelerate chemical reactions and are essential for cell function. They are almost always proteins, and their proper three-dimensional structure allows their biological activity. The activity of enzymes can be inhibited by various inhibitors, which many drugs are, and many enzymes are also subject to active drug development.
As a result of activation of the hydroxysteroid-17-beta dehydrogenase type 10 (HSD17β10) gene, HSD17β10 enzyme, also known as type II 3-hydroxyacyl-CoA dehydrogenase, is produced. This mitochondrial enzyme induces the oxidation of fatty acids, alcohols, and steroids. It catalyzes, for example, the oxidation of a biologically active estrogen, 17β-estradiol, to a less active estrone, the oxidation of 3α-androstanediol to 5α-dihydrotestosterone, and the oxidation of allopregnanolone to 5α-dihydroprogesterone.
HSD17β10 enzyme activity has been observed in different regions of the brain, most in the hippocampus. In patients with Alzheimer's disease, levels of the enzyme HSD17β10 increase, resulting in a decrease in the levels of estradiol and allopregnanolone, which are known for their protective role in the nervous system. By inhibiting the HSD17β10 enzyme it is possible to maintain the neuroprotective steroid levels of the brain, which is why this enzyme is an interesting target for drug development.
The goal of the thesis was to develop a new activity test for the enzyme HSD17β10, which could later be used in screening drug candidates. During the development of the method, scientific publications and in-house experience in the development of similar enzyme activity tests were utilized. The work included analyses of various factors influencing the activity of the enzyme. These analyses were carried out in the 96 well plate format using fluorescence and UPLC methods.
The objectives set for the thesis were realized as a new activity test for the HSD17β10 enzyme was successfully developed as part of the company's drug screening.
