Tiivistelmä
Trichoderma reesei -hometta voidaan hyödyntää teollisuudessa entsyymien tuotto-organismina, ja se voidaan geneettisesti muuntelemalla saada tuottamaan myös erilaisia heterologisia tai homologisia rekombinanttiproteiineja. Sen luontaisesti tuottamia sellulaaseja ja hemisellulaaseja voidaan hyödyntää esimerkiksi toisen sukupolven bioetanolin tuotantoprosesseissa, mutta lignosellulolyyttisten entsyymien muodostamat kustannukset ovat pysyneet näiden prosessien osalta varsin korkeina. Tehokkaampien tuottokantojen muodostaminen ja uusien edullisempien entsyymien tuottoon soveltuvien raaka-aineiden löytäminen voivat auttaa alentamaan toisen sukupolven bioetanolin tuotantoprosesseissa tarvittavien entsyymien muodostamia kustannuksia. Tämän opinnäytetyön tavoitteena oli selvittää erityisten geneettisesti muunneltujen Trichoderma reesei -kantojen entsyymituottopotentiaalia erilaisia raaka-aineita hyödyntäen.
Työssä käytettiin kahta erilaista geneettisesti muunneltua kantatyyppiä, joista toinen edusti isäntäkantana perinteisempää kantalinjaa, jotka tyypillisesti tarvitsevat erityiset indusoivat raaka-aineet entsyymien tuottamiseksi. Toisella kantatyypillä voitiin tuottaa entsyymejä myös glukoosipohjaista tuottoprosessia hyödyntäen, mikä ei ole ollut perinteisillä kannoilla normaalisti mahdollista. Kokeet suoritettiin bioreaktoreissa hallituissa olosuhteissa panossyöttöprosesseina. Bioreaktorikasvatuksista otetuista näytteistä mitattiin näytteiden kiintoainepitoisuus, tiettyjen liukoisten substraattien määrä sekä kasvuliuokseen eritettyjen proteiinien kokonaismäärä. Käsitys kantojen tuottamasta entsyymiprofiilista ja niiden keskinäisistä eroavaisuuksista muodostettiin analysoimalla kantojen kasvuliuokseen tuottamia proteiineja SDS-PAGE-geelien avulla.
Tulosten perusteella käytetyillä raaka-aineilla saatiin tuotettua entsyymejä, mutta tuotetut proteiinimäärät, saannot sekä tuottavuudet vaihtelivat suuresti kantatyyppien ja eri raaka-aineiden välillä. SDS-PAGE-geelien analyysin perusteella tuotetuista entsyymiprofiileista nähtiin eroavaisuuksia eri kantojen ja raaka-aineiden välillä, mutta selkein eroavaisuus vaikutti olevan glukoosipohjaisessa tuottoprosessissa, jossa käytetyt kannat näyttivät tuottaneen ksylanaasi I ja ksylanaasi II entsyymejä isäntäkantoja enemmän. Työssä käytettyjen tuottoprosessien osalta tarvittaisiin kuitenkin vielä optimointia, jotta saadaan tietoa johtuvatko eroavaisuudet tuloksissa esimerkiksi käytettyjen raaka-aineiden lisäksi myös niiden keskinäisestä määrästä tai käytetyistä syöttönopeuksista, vai onko kyseessä enemmän tuottokantoihin liittyvät rajoittavat ominaisuudet. The filamentous fungus Trichoderma reesei can be used as a production organism in industrial scale enzyme production and it can also be genetically engineered to produce various types of heterologous or homologous recombinant proteins. Cellulases and hemicellulases it produces naturally can be utilized for example in second-generation bioethanol production processes but the cost of lignocellulolytic enzymes needed in these processes has remained relatively high. The development of more efficient production hosts and discovery of new low-cost feedstock materials can help to reduce the enzyme costs in second generation bioethanol production processes. The aim of this thesis was to examine enzyme production potential of specific genetically engineered Trichoderma reesei strains using various feedstocks.
Two genetically engineered strain types were used. The first represented more conventional strain type as a host strain which usually requires specific inducing feedstocks to produce enzymes. The second strain type enabled enzyme production using a glucose-based production process which has not been normally possible with conventional strain types. The experiments were carried out in bioreactors under controlled conditions as fed-batch processes. Solid’s content, the amount of certain soluble substrates and the total amount of proteins secreted into the growth medium, were measured from the cultivation samples. An understanding of the enzyme profile produced by the strains and the differences between them was obtained by SDS-PAGE analysis of the proteins secreted into the growth medium.
The results indicate that enzymes could be produced with the tested feedstocks, but the total amount of protein produced, the yields and the productivities varied greatly between the strain types and different feedstocks. SDS-PAGE analysis showed differences in the produced enzyme profiles between the used strain types and feedstocks but the clearest difference seemed to be in the glucose-based production process where the strains appeared to produce more xylanase I and xylanase II enzymes than the host strains. However, further optimization would be needed regarding the used processes to understand whether the differences in results are due not only to the used feedstock materials but also to the quantities or used feed rates or whether they are more due to the limiting characteristics of the strains.
