Tiivistelmä
Opinnäytetyön tarkoituksena oli optimoida Wallac Oy:n valmistamien Anti-IRT mikrotiitterilevyjen tuotantoprosessi niin, että olemassa olevat resurssit voidaan hyödyntää tehokkaasti ja tuotteen kysyntään voidaan vastata paremmin. Työn tavoitteena oli kartoittaa, voidaanko levyvalmistuksessa valmistaa vasta-aineella pinnoitettuja Anti-IRT mikrotiitterilevyjä pidemmällä inkubointiajalla sekä suuremmalla eräkoolla siten, että tuotteen laadunvalvonta- ja säilyvyyskriteerit täyttyvät.
Opinnäytetyön kirjallinen osuus käsittelee lääkinnällisten laitteiden kansallista sekä kansainvälistä säännöstelyä, Wallac Oy:n sisäisiä järjestelmiä, immunomääritysteknologiaa sekä immunoreaktiivisen trypsinogeenin (IRT) ja kystisen fibroosin seulonnan merkitystä.
Eräkoon kasvatuksen myötä tuotantoprosessin kesto pidentyy, minkä vuoksi karakterisoinnissa oli kriittistä selvittää, pysyykö mikrotiitterilevyille annosteltava vasta-aineliuos tasalaatuisena pidemmällä annosteluajalla. Karakterisointia varten valmistettiin kaksi pientä testierää kahdella annosteluajalla. Pidentämällä prosessissa käytettyä inkubointiaikaa, tuotannonsuunnitteluun luodaan joustavuutta sekä varmistetaan tuotteen valmistettavuus esimerkiksi tuotantolinjan pitkittyneessä vikatilanteessa. Inkubointiaikaa karakterisoitiin kolmella aikapisteellä, joille valmistettiin pienet testierät.
Karakterisoinnin tuloksien perusteella Anti-IRT mikrotiitterilevyeriä voidaan tuottaa pidennetyllä inkubointiajalla ja kasvatetulla eräkoolla, siten että vasta-aineella pinnoitetut mikrotiitterilevyt täyttävät laadunvalvonta- sekä säilyvyyskriteerit. Opinnäytetyön tuloksien perusteella Anti-IRT mikrotiitterilevyille voidaan aloittaa verifiointiprosessi suuremman eräkoon sekä pidemmän inkubointiajan käyttöönottamiseksi. The purpose of the thesis was to optimize the production process of Anti-IRT microtitration plates manufactured by Wallac Oy, so that existing resources can be utilized efficiently and product demand could be better met. The aim of this thesis was to research whether it is possible to manufacture antibody-coated Anti-IRT microtitration plates in coated plate manufacturing with longer incubation time as well as larger batch size so that the quality control and shelf life criteria of the product are fulfilled.
The theoretical basis of this thesis project covers the national and international regulation of medical devices, the internal systems of Wallac Oy, immunoassay technology, and the significance of immunoreactive trypsinogen (IRT) and cystic fibrosis screening.
With the increase in batch size, the duration of the production process is extended, which is why it was critical in characterization to determine whether the antibody solution administered to microtiter plates remains homogeneous over longer dosing time. For batch size characterization, two small batches were manufactured with two dosing times. By extending the incubation time used in the process, flexibility is created in the production planning and the producibility of the product is ensured even in a prolonged production machinery failure situation. The incubation time was characterized by three time points for which small test batches were prepared.
Based on the results of the characterization, Anti-IRT microtiter plate batches can be produced with extended incubation time and increased batch size, so that antibody-coated microtitration plates meet quality control and shelf life criteria. Based on the results of the thesis, a verification process can be initiated for Anti-IRT microtitration plates to introduce a larger batch size as well as the longer incubation time.
