Tiivistelmä
Uusien lääkeaineiden kehitystyötä hankaloittaa usein lääkeaineiden niukkaliukoisuus. Niukkaliukoisuus johtuu lääkeaineiden kiderakenteesta ja se hidastaa ja hankaloittaa lääkeaineiden läpäisevyyttä kehon kalvorakenteissa. Liukoisuusominaisuuksia voidaan parantaa saattamalla lääkeaineet amorfiseen muotoon eli olomuotoon, jolla ei ole toistuvaa, järjestäytynyttä kiderakennetta. Amorfinen muoto on kuitenkin epästabiili, ja uudelleenkiteytymistä voi alkaa tapahtua jo valmistuksen tai säilytyksen aikana. Amorfisuutta on mahdollista ylläpitää esimerkiksi kantajaväliaineen eli matriisin avulla.
Tämän työn tavoitteena oli muodostaa lääkeaineesta ja matriisista amorfinen formulaatio, ja tutkia formulaatiossa käytetyn selluloosapohjaisen matriisimateriaalin kykyä säilyttää lääkeaine amorfisessa muodossaan ajan kuluessa. Lisäksi työssä tutkittiin lääkeaineen vapautumista matriisista.
Opinnäytetyön tuloksena voitiin todeta, että käytetty matriisi säilytti lääkeaineen amorfisuuden koko työn ajan. Lääkeaineen vapautuminen matriisista vaatii tulevaisuudessa formulaation edelleenkehitystä. The development of new drugs is often hampered by the drug’s low solubility in water. Low solubility is caused by the crystal structure of the drug and slows down and complicates the permeability of the drug in the membrane structures of the body. Solubility can be improved by converting the drugs into amorphous form, where they have no repetitive, orderly crystal structure. However, the amorphous form is unstable, and recrystallization may occur already during manufacture or storage. It is possible to maintain amorphism for example by using a matrix.
The aim of this thesis project was to form an amorphous formulation of the drug and matrix, and to study the ability of the cellulose-based matrix material used in the formulation to retain the drug in its amorphous form over time. In addition, the release of the drug from the matrix was studied in the work.
As a result of the thesis, it could be noted that the matrix used maintained the amorphism of the drug throughout the work. The release of the drug from the matrix requires further development of the formulation in the future.
