Tiivistelmä
Tässä alustavassa farmakokineettisessä in vivo -hiiritutkimuksessa etsittiin vastausta kahteen kysymykseen; Vaikuttaako hiirten käsittely suun kautta annostellun tunnetun lääkeaineen imeytymisen parametreihin ja onko 10 päivän treenaukseen kuluva työaika perusteltu lisä tutkimusten hinnoittelussa. Vertaillut käsittelyryhmät olivat hännästä nostellut, tuubilla nostellut ja treenatut hiiret. Lääkeaineen imeytymisen oletettiin viivästyvän stressin vaikutuksesta. Treenin hyötyjä tarkasteltiin kipuilmeiden pisteytyksellä, jossa ryhmien keskiarvopisteiden oletettiin olevan samat käsittelystä riippumatta.
Lääkealtistuksen parametrit kvantitoitiin sopivalla bioanalyyttisellä menetelmällä ja laskettiin ei-osastoanalyysillä. Hiirten treenauksen hyötyjä pohdittiin farmakokineettisten tulosten sekä eri ryhmien kokeman kivun eron perusteella. Kasvojen ilmeiden ryhmävertailu suoritettiin tilastollisesti Kruskal-Wallis-testillä.
Treenattujen hiirien lääkealtistuksen parametrit olivat lupaavimmat. 10 päivän treeni todettiin mahdottomaksi toteuttaa käytännössä. Hännästä nosteltujen hiirien kipuilmeiden pisteytyksessä oli merkittävä ero verrattuna putkella nosteltuihin hiiriin. Kahden ei-aversiivisen ryhmän, putkella nostettujen ja treenattujen hiirten, välillä ei ollut merkittävää eroa kasvojen pisteytyksessä.
Hännästä nostelua pidetään jatkossa vältettävänä käsittelymenetelmänä. Hiirten treenauksella on positiivista vaikutusta tutkitun lääkeaineen imeytymisen farmakokinetiikkaan. Kolmen päivän treenin hyötyjä on tutkittava lisää. The present preliminary pharmacokinetic in vivo mouse study sought the answer to the following two questions: Does the handling of mice influence the absorption parameters of an orally administered known drug and is the 10 days of work time spent on training the mice justified from the pricing point of view. The compared handling groups were tail lifted, tube lifted and trained. The stress caused by the handling was assumed to delay the absorption of the drug. The stress during the procedures was examined by scoring facial pain markers and the mean scores of the groups were assumed to be the same regardless of the handling.
The parameters of drug exposure were quantified with an appropriate bioanalytical method and calculated with the non-compartmental analysis. The benefits of mice training are discussed based on the pharmacokinetic results and the difference in pain experienced by different groups. The group comparisons of the facial expressions were performed using the Kruskal-Wallis test.
The parameters of the drug exposure of the trained mice were the most promising. The 10-day training was found to be impossible to carry out in practice. There was a significant difference in the facial expression scores of the tail lifted mice compared with the tube lifted mice. Between the two non-aversive groups, tube lifted and trained mice, there were no significant difference in the facial scores.
As a result of the study, tail lifting will be considered as a handling method to be avoided in the future. Training mice had a positive effect on the pharmacokinetics of the studied drug absorption. The benefits of more practical three days of training needs to be studied more.
